引言
为提高化学品对鱼类生物蓄积作用的预测能力,需建立鱼体内肝脏生物转化的评估方法并将相关
信息纳入已建立的计算机模型。目前主要利用肝组织的体外代谢系统测定化学品的固有清除率。
本文件描述了基于虹鳟鱼(Oncorhynchusmykiss)肝S9亚细胞组分(RT⁃S9),采用底物清除法测定
受试物体外固有清除率()的试验方法。OECD化学品测试导则No.319A和OECD化学
CLINVITRO,INT
品测试指南文件No.280描述了开展试验的详细步骤,以及如何将用于鱼类生物蓄积性的
CLINVITRO,INT
计算机模型预测。
简而言之,根据确定的估算肝脏固有清除率,并采用基于生理学的毒代动力学
CLINVITRO,INT
(PBTK)模型进一步评估鱼类的生物蓄积作用。也可采用合适的体外-体内外推模型(IVIVE)将该值
外推获得全身(体内)的生物转化速率常数。如果受试物处于模型的应用范围内,可将测得的
作为输入信息,应用于基于生物转化的生物富集计算机预测模型。
CLINVITRO,INT
体内生物转化率可用于计算机模型预测生物富集系数(BCF)。将体外方法获得的生物转化数据
整合到鱼类一室模型中,可显著改善模型性能,与代谢缺失时得到的预测结果相比,整合生物转化数据
后的模型对生物富集结果的预测更接近试验值。
本文件的重复性已通过6个实验室开展的比对试验(针对5种受试物和1种参比物)进行了验证。
