1范围
本文件描述了饲用果胶酶活力的分光光度测定方法。
本文件适用于饲料添加剂果胶酶和含有果胶酶的混合型饲料添加剂酶制剂中果胶酶活力的测定。
本文件不适用于碱性果胶酶活力的测定。
本方法定量限为10U/g(或U/mL)。

3术语和定义
下列术语和定义适用于本文件。
3.1
果胶酶活力单位pectinaseactivityunit
在37℃、pH3.5的条件下,每小时从质量浓度为2.25mg/mL的果胶溶液中分解释放1mg还原糖所
需要的酶量。
注:以U表示。
4原理
果胶酶水解果胶溶液产生还原糖,用3,5﹘二硝基水杨酸显色,以分光光度计比色测定,计算果胶
酶活力。
5试剂或材料
除非另有说明,仅使用分析纯试剂。
5.1水:GB/T6682,三级。
5.2氢氧化钠溶液(200g/L):称取氢氧化钠20.0g,加水溶解,冷却至室温后定容至100mL。
5.3柠檬酸溶液(0.1mol/L):称取一水柠檬酸21.01g,加水溶解并定容至1000mL。
5.4柠檬酸三钠溶液(0.1mol/L):称取二水合柠檬酸三钠29.41g,加水溶解并定容至1000mL。
5.5柠檬酸﹘柠檬酸钠缓冲液(0.1mol/L,pH=3.5):取柠檬酸溶液(5.3)700mL,柠檬酸三钠溶
液(5.4)300mL,混匀,用柠檬酸溶液(5.3)或柠檬酸三钠溶液(5.4)调节pH至3.5。
5.6果胶溶液(3.0mg/mL):称取果胶(≥74%聚半乳糖醛酸,CAS号:9000﹘69﹘5)0.3g,精确
